Assessment of polycyclic aromatic hydrocarbon concentrations in mussels (Mytilus galloprovincialis) from the Western basin of the Mediterranean Sea ArchiMer
Galgani, Francois; Martinez-gomez, Conception; Giovanardi, Franco; Romanelli, Giulia; Caixach, Josep; Cento, Alessandro; Scarpato, Alfonso; Benbrahim, Samir; Messaoudi, Sabri; Deudero, Salud; Boulahdid, Mostafa; Benedicto, Jose; Andral, Bruno.
The Mytilos project was carried during the last 3 years (2004-2006) in order to evaluate the level of chemical contamination along the coasts of the western basin of the Mediterranean Sea. Active mussel watch consisted of caged (12 weeks) Mytilus galloprovincialis at 123 stations along the Mediterranean coasts of Spain, France, Italy, North Tunisia, Algeria and Morocco. Results indicated total PAHs levels in the range 22-106 mu g/kg dry weight. All large towns or industrial areas from all countries of the basin were affected. Analysis of the origin of contamination demonstrated mainly a pyrolytic origin except in some areas such as Maddalena, Napoli, Alger, and Cornigliano (Genova). Statistical analysis of the PAHs accumulation data enabled the...

Comparison of Polycyclic Aromatic Hydrocarbon (PAHs) concentrations in urban and natural forest soils in the Atlantic Forest (São Paulo State) Anais da ABC (AABC)
Bourotte,Christine; Forti,Maria Cristina; Lucas,Yves; Melfi,Adolpho J..
Studies about pollution by Polycyclic Aromatic Hydrocarbons (PAHs) in tropical soils and Brazil are scarce. A study was performed to examine the PAHs composition, concentrations and sources in red-yellow Oxisols of remnant Atlantic Forest of the São Paulo State. Sampling areas were located in an urban site (PEFI) and in a natural one (CUNHA).The granulometric composition, pH, organic matter content and mineralogical composition were determined in samples of superficial soils. The sum of PAHs (ΣHPAs) was 4.5 times higher in the urban area than in the natural one. Acenaphthylene, acenaphthene, fluorene, phenanthrene and fluoranthene have been detected in the soils of both areas and presented similar concentrations. Acenaphthene and fluorene were the most...

Contamination chimique du milieu marin : de la mesure à l'évaluation des risques ArchiMer
Abarnou, Alain.
This activity report entitled (“Chemical contamination of the marine environment: from measurement to risk assessment”) summarizes the main stages my career at Ifremer from November 1975 to june 2013. The first chapter deals with the seawater chlorination and chlorinated waters released into the sea, the identification and the conditions of the formation of various by-product of these processes. The second chapter concerns PCBs, their nature, their chemical analysis in environmental matrices. PCBs are considered as the best examples of persistant and bioaccumulable compounds and therefore, in all this report they are flagship contaminants to which other groups of studied substances are compared. The third chapter refers to studies performed within marine...

Development of a reference artificial sediment for chemical testing adapted to the MELA sediment contact assay ArchiMer
Le Bihanic, Florane; Perrichon, Prescilla; Landi, Laure; Clerandeau, Christelle; Le Menach, Karyn; Budzinski, Helene; Cousin, Xavier; Cachot, Jerome.
Most persistent organic pollutants, due to their hydrophobic properties, accumulate in aquatic sediments and represent a high risk for sediment quality. To assess the toxicity of hydrophobic pollutants, a novel approach was recently proposed as an alternative to replace, refine and reduce animal experimentation: the medaka embryo–larval sediment contact assay (MELAc). This assay is performed with Japanese medaka embryos incubated on a natural sediment spiked with the compound being tested. With the aim of improving this assay, our study developed a reference exposure protocol with an artificial sediment specifically designed to limit natural sediment composition uncertainties and preparation variability. The optimum composition of the new artificial...

Effects of 16 pure hydrocarbons and two oils on haemocyte and haemolymphatic parameters in the Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer
Bado Nilles, A; Gagnaire, Beatrice; Thomas-guyon, Helene; Le Floch, S; Renault, Tristan.
The in vitro effects of polycyclic aromatic hydrocarbons (PAHs) on haemocyte and haemolymphatic parameters of the Pacific oyster, Crassostrea gigas, were tested using field concentrations (10(-7) and 10(-9) mg mL(-1)) observed in the Marennes-Oleron Basin (Charente-Maritime, France) and high concentrations (10(-3) and 10(-5) mg mL(-1)) observed during oil spills. As a first step, the effects of pollutants, after a 24 h contamination period, were monitored on individual and pooled haemolymph samples and similar results were observed for both sample types. In a second step, haemolymphs from 45 oysters were withdrawn and pooled. Eighteen pollutants were tested in vitro after a 4 and 24 h contamination period and 10 of them showed significant modulations of...

Influence of sediment composition on PAH toxicity using zebrafish (Danio rerio) and Japanese medaka (Oryzias latipes) embryo-larval assays ArchiMer
Perrichon, Prescilla; Le Bihanic, Florane; Bustamante, Paco; Le Menach, Karyn; Budzinski, Helene; Cachot, Jerome; Cousin, Xavier.
Due to hydrophobic and persistent properties, polycyclic aromatic hydrocarbons (PAHs) have a high capacity to accumulate in sediment. Sediment quality criteria, for the assessment of habitat quality and risk for aquatic life, include understanding the fate and effects of PAHs. In the context of European regulation (REACH and Water Framework Directive), the first objective was to assess the influence of sediment composition on the toxicity of two model PAHs, benzo[a]pyrene and fluoranthene using 10-day zebrafish embryo-larval assay. This procedure was undertaken with an artificial sediment in order to limit natural sediment variability. A suitable sediment composition might be then validated for zebrafish and proposed in a new OECD guideline for chemicals...

L'utilisation des échantillonneurs passifs. Une nouvelle méthode pour évaluer la contamination chimique des masses d'eau ArchiMer
Gourlay, Catherine; Gonzalez, Jean-louis.
The Water European Framework Directive requires the implementation of programs to monitor the chemical quality of water bodies (freshwater, transition water and seawater), based on grab measurements of concentrations of various contaminants (metals, organic compounds). The results are limited by several analytical difficulties, and the poor representativity because of the temporal variability of contamination. The passive sampling techniques, which basically consist of concentrating substances on a submerged device for a given period, and analyzing accumulated substances, should improve the monitoring, by simplifying analytical issues (lower detection limits, analysis in a simpler matrix), and allowing time integration of the contamination. Here, we...

Active bio-monitoring in terms of biornarkers was attempted using Crassostrea gigas larvae produced in the laboratory and transplanted using special containers to two sites at the entrance (A) and inner part (P) of the harbour of Arcachon (French Atlantic Coast). The larvae were kept in the medium for 48 h. Their physiological status and their biomarker levels : acetylcholinesterase AChE, catalase CAT and glutathione S-transferase GST activities were determined together with metallothionein Wand Thiobarbituric Acid Reactive Substances TBARS concentrations. Copper and PAH (polycyclic aromatic hydrocarbon) concentrations were determined in the exposed larvae and in the sediments collected under the containers. Cadmium, lead and zinc could be also analyzed in...

New SERS Substrates For Polycyclic Aromatic Hydrocarbon (PAH) Detection: Towards Quantitative SERS Sensors For Environmental Analysis ArchiMer
Peron, Olivier; Rinnert, Emmanuel; Compere, Chantal; Toury, T.; De La Chapelle, M. Lamy.
In the investigation of chemical pollutions, such as PAHs (Polycyclic Aromatic Hydrocarbons) at low concentration in aqueous medium, surface-enhanced Raman scattering (SERS) stands for an alternative to the inherent low cross-section of normal Raman scattering. Indeed, SERS is a very sensitive spectroscopic technique due to the excitation of the surface plasmon modes of the nanostructured metallic film.

Organic micropollutants (PAHs, PCBs, pesticides) in seaweeds of the lagoon of Venice ArchiMer
Pavoni, B; Caliceti, M; Sperni, L; Sfriso, A.
Samples of seven genera of seaweeds (Ulva, Gracilaria, Porphyra, Grateloupia, Undaria, Fucus and Cystoseira), which can be used as environmental biomonitors and in several applications like human alimentation, animal feeding and cosmetics, were collected in four sampling sites in the Lagoon of Venice in spring and autumn 1999 with the aim of determining the contamination due to organic micropollutants (polychlorinated biphenyls. PCBs; chlorinated pesticides; polycyclic aromatic hydrocarbons, PAHs). On an average, most of the considered species were contaminated especially by PAHs (up to 56 ng g(-1) dry wt. (dw)) whereas concentrations of PCBs and pesticides in macroalgal tissues were significantly correlated and did not exceed 5 ng g-1 dw. Among the...

Pollutant effects on Pacific oyster, Crassostrea gigas (Thunberg), hemocytes: Screening of 23 molecules using flow cytometry ArchiMer
Gagnaire, Beatrice; Thomas Guyon, H; Burgeot, Thierry; Renault, Tristan.
The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Oysters may be suitable for studies on disturbance by pollutants of physiological activities, among which defense mechanisms are poorly documented in bivalves. In this study, effects of pollutants on hemocyte functions were monitored in Pacific oyster, C. gigas. Hemocytes were exposed...

Polycyclic aromatic hydrocarbons degradation by marine-derived basidiomycetes: optimization of the degradation process BJM
Vieira,Gabriela A.L.; Magrini,Mariana Juventina; Bonugli-Santos,Rafaella C.; Rodrigues,Marili V.N.; Sette,Lara D..
ABSTRACT Pyrene and benzo[a]pyrene (BaP) are high molecular weight polycyclic aromatic hydrocarbons (PAHs) recalcitrant to microbial attack. Although studies related to the microbial degradation of PAHs have been carried out in the last decades, little is known about degradation of these environmental pollutants by fungi from marine origin. Therefore, this study aimed to select one PAHs degrader among three marine-derived basidiomycete fungi and to study its pyrene detoxification/degradation. Marasmiellus sp. CBMAI 1062 showed higher levels of pyrene and BaP degradation and was subjected to studies related to pyrene degradation optimization using experimental design, acute toxicity, organic carbon removal (TOC), and metabolite evaluation. The experimental...

Responses of the European flounder (Platichthys flesus) to a mixture of PAHs and PCBs in experimental conditions ArchiMer
Dupuy, Celie; Galland, Claire; Devaux, Alain; Bony, Sylvie; Loizeau, Veronique; Danion, Morgane; Pichereau, Vianney; Fournier, Michel; Laroche, Jean.
A multibiomarker approach was developed to evaluate the juvenile European flounder responses to a complex mixture of 9 polycyclic aromatic hydrocarbons (PAHs) and 12 polychlorinated biphenyls (PCBs). Exposure was performed through contaminated food pellets displaying: (1) PAH and PCB levels similar to those detected in the heavily polluted Seine estuary, respectively in sediments and in flatfish and (2) ten times these concentrations. Several biomarkers of the immune system (e.g., lysozyme concentration and gene expression of complement component C3 and TNF-receptor), DNA damage (e.g., Comet assay), energetic metabolism (e.g., activity of cytochrome C oxidase), detoxification process (e.g., cytochrome P450 1A1 expression level: CYP1A1; betaine homocysteine...

Seasonal changes of chemical contamination in coastal waters during sediment resuspension ArchiMer
Bancon-montigny, Chrystelle; Gonzalez, Catherine; Delpoux, Sophie; Avenzac, Muriel; Spinelli, Sylvie; Mhadhbi, Takoua; Mejri, Kaouther; Hlaili, Asma Sakka; Pringault, Olivier.
The potential of remobilization of pollutants is a major problem for anthropogenic ecosystems, because even when the anthropogenic source of pollution is identified and removed, pollutants stored in sediments can be released into the water column and impact pelagic communities during sediment resuspension provoked by dredging, storms or bottom trawling. The objectives of the present study were to assess the changes observed in the chemical composition of the water column following resuspension of a polluted marine sediment and the consequences for the chemical composition of adjacent marine waters according to season. For that purpose, an experimental sediment resuspension protocol was performed on four distinct occasions, spring, summer, fall and winter,...

he objective of this study is to demonstrate the successful functionalization of the surface of a chalcogenide infrared waveguide with the ultimate goal of developing an infrared micro-sensor device. First, a polyisobutylene coating was selected by testing its physico-chemical compatibility with a Ge-Sb-Se selenide surface. To simulate the chalcogenide platform infrared sensor, the detection of benzene, toluene, and ortho-, meta- and para-xylenes was efficaciously performed using a polyisobutylene layer spin-coated on 1 and 2.5 µm co-sputtered selenide films of Ge28Sb12Se60 composition deposited on a zinc selenide prism used for attenuated total reflection spectroscopy. The thickness of the polymer coating was optimized by attenuated total reflection...

Trace metals and PAHs in topsoils of the University campus in the megacity of São Paulo, Brazil Anais da ABC (AABC)
BOUROTTE,CHRISTINE L.M.; SUGAUARA,LUCY E.; MARCHI,MARY R.R. DE; SOUTO-OLIVEIRA,CARLOS E..
Abstract: The aim of this study is to discuss the concentration distribution, composition and possible sources of trace metals and 13 PAHs in topsoils of the University campus, in the city of São Paulo, the largest city of South America. Mineralogy and granulometry of topsoils (0-10 cm) samples, were determined and As, Ba, Cd, Co, Cr, Cu, Mo, Ni, Pb, Sb, V, Zn, Hg, Pt, Pd and PAHs concentrations were quantified in the bulk fraction. The average clay content of soils was 221±61.8 g kg-1 and total carbon was low (mean of 25 g kg-1). Concentrations of metals were generally lower than the reference value established by the São Paulo State Environmental Agency and other studies in the São Paulo urban area, except for Cd, Ni, Pb, Cr and Zn. Use of clearance rate in Choromytilus chorus (Bivalvia: Mytilidae) as a non-destructive biomarker of aquatic pollution RChHN
TORO,BEATRIZ; NAVARRO,JORGE M.; PALMA-FLEMING,HERNÁN.
Reduction in clearance rate was observed in the mussel Choromytilus chorus in relation to the degree of environmental pollution. Three sampling sites included the polluted San Vicente Bay, middling polluted Corral Bay, and weakly polluted Yaldad Bay in southern Chile. Pollution levels were estimated by quantitative analysis of polynuclear aromatic hydrocarbons (PAHs) in mussel tissue from the three sites. Mussels from San Vicente showed lowest clearance rates and highest PAHs levels in both spring 1998 and summer 1999. Populations from Corral and Yaldad showed generally low levels of PAHs, so the mussels from Yaldad showed the highest rates of clearance in comparison to Corral and San Vicente populations. The significant negative relationship (P <...

Use of low density polyethylene membranes for assessment of genotoxicity of PAHs in the Seine River ArchiMer
Vincent-hubert, Francoise; Uher, Emmanuelle; Di Giorgio, Carole; Michel, Cecile; De Meo, Michel; Gourlay-france, Catherine.
The genotoxicity of river water dissolved contaminants is usually estimated after grab sampling of river water. Water contamination can now be obtained with passive samplers that allow a time-integrated sampling of contaminants. Since it was verified that low density polyethylene membranes (LDPE) accumulate labile hydrophobic compounds, their use was proposed as a passive sampler. This study was designed to test the applicability of passive sampling for combined chemical and genotoxicity measurements. The LDPE extracts were tested with the umu test (TA1535/pSK1002 ± S9) and the Ames assay (TA98, TA100 and YG1041 ± S9). We describe here this new protocol and its application in two field studies on four sites of the Seine River. Field LDPE extracts were...
